Legundi leaves (Vitex trifolia L.) have been used to treat asthma as traditional practices, and temulawak (Curcuma xanthorrhiza Roxb.) have known as antiinflamation agent. Combination both of them suspected have better effect to treat bronkial asthma caused by alergen. Purposed of this experiment is knowing inhibition contraction of isolation guinea pig trachea induced by histamine from combination ethanolic extract legundi leaves and temulawak.
Methods of this experiment in vitro, using trachea isolation from guinea pig (Cavia parcellus) male depend in Krebs solution. The doses combination extract etanolic legundi leaves and temulawak are 100:0, 75:25, 50:50, 25:75, and 0:100. The dose for 100 part extract is 0,5 mg/ml in organbath. Difenhidramin as antihistaminic, use as positif control. Trachea smooth muscle induced by cumulatif dose of histamine dihidroklorida as the agonist. Trachea contraction measuring by pharmacodinamic equipment. Then changed to contraction response (%) and inhibition contraction (%).
Statictic analyse use LSD (least significant differences) level 95% showed differences between each group and negative control. Inhibition induced by 1.10-4 M histamine dihidroklorida used combination extract etanolic legundi leaves and temulawak 100:0, 75:25, 50:50, 25:75, and 0:100 are 70,43±2,59, 58,14±3,04, 59,70±3,04, 46,80±4,84 and 40,61±1,51%. LSD analyse inhibitie contraction result that composition combination 100:0, 75:25, dan 50:50 showed no differences between difenhidramin as positif control while LSD result to compare 75:25 and 50:50 between 100 part of legundi leaves ethanolic extract showed no differences.
